Topic description
Nos équipes (MoNALISA-SPCMIB et DDR-IPBS) ont récemment découvert, à travers une approche multidisciplinaire en collaboration avec le LCC, le mécanisme d'action cytotoxique d'une vaste famille de lipides acétyléniques bioinspirés. Ces molécules agissent comme des prodrogues énantiospécifiquement bioactivées par une Short-Chain Dehydrogenase/Reductase (SDR) en des espèces cytotoxiques hautement électrophiles. Afin d'exploiter le potentiel thérapeutique de cette découverte contre le cancer du poumon à petites cellules, nous avons développé de nouvelles approches de synthèse permettant de générer des dérivés plus actifs, plus sélectifs et plus stables dans les milieux biologiques et chez l'animal. De plus, nous avons montré que ce mécanisme de bioactivation est généralisable, c'est à dire que de nouvelles prodrogues sélectivement activées par d'autres SDRs peuvent être développées.
Nous sommes actuellement les seules équipes à exploiter la bioactivation par les SDRs pour des approches pharmacologiques innovantes. Le projet que nous proposons vise à capitaliser sur nos avancées pour développer les premières prodrogues bioactivées par deux SDRs surexprimées dans des cancers de mauvais pronostic, en particulier les cancers du foie et du pancréas, mais d'expression très restreinte dans les tissus normaux. Nous avons déjà identifié des composés hits pour chacune des deux enzymes. Le/la candidat.e chimiste (encadrement MoNALISA-SPCMIB) exploitera une nouvelle approche modulaire de synthèse développées en collaboration avec le LCC pour générer un panel d'analogues avec l'objectif d'augmenter l'activité, la sélectivité et la stabilité dans l'animal. Un(e) doctorant(e) biologiste (encadrement DDR-IPBS) testera ces analogues, validera leur mécanisme d'action et leur activité anticancéreuse sur des modèles murins. Ces travaux permettront de développer de nouvelles thérapies ciblées contre le cancer, tout en renforçant la position de leader de Toulouse sur ces nouvelles molécules thérapeutiques.

In order to exploit the therapeutic potential of this discovery against small cell lung cancer, we have developed new synthetic approaches to generate derivatives that are more active, more selective and more stable in biological environments and in animals. Furthermore, we have shown that this bioactivation mechanism is generalizable, that is to say that new prodrugs selectively activated by other SDRs can be developed.
We are currently the only teams to exploit bioactivation by SDRs for innovative pharmacological approaches. A PhD biologist student (DDR-IPBS supervision) will test these analogues, validate their mechanism of action and their anticancer activity on mouse models. This work will make it possible to develop innovative targeted therapies against cancer, while strengthening Toulouse's leadership in these new therapeutic avenues.

Candidate's profile
Le/la candidat.e doit avoir une très solide formation en chimie organique avec des connaissances avancées en pharmacochimie et chimie thérapeutique. Le/la candidat.e devra maîtriser complétement les concepts et outils de la synthèse organique multi-étapes avec de solides compétences théoriques et pratiques en stéréochimie, réactivité, purification et caractérisation structurale de petites molécules. Des notions de conception in silico de molécules assistée par docking seraient souhaitables. Le/la candidat.e doit aussi être disposé.e à intégrer un environnement de travail hautement interdisciplinaire en étroit partenariat avec une équipe de biologistes. Des notions théoriques et pratiques en biologie cellulaire et moléculaire seront un atout.
The candidate must have very solid training in organic chemistry with advanced knowledge in pharmacochemistry and therapeutic chemistry. The candidate must completely master the concepts and tools of multi-step organic synthesis with solid theoretical and practical skills in stereochemistry, reactivity, purification and structural characterization of small molecules.
