Description du sujet
Les protéinopathies, comme les maladies d'Alzheimer, de Huntington et de Parkinson, sont des maladies neurodégénératives dues à des protéines mal repliées qui s'agrègent dans le cerveau, acquérant des fonctions toxiques et recrutant d'autres protéines solubles. Le vieillissement de la population accroît l'importance de détecter et de suivre l'agrégation de ces protéines de manière non invasive pour les diagnostics et la recherche. La méthode d'IRM-CEST (Chemical Exchange Saturation Transfer) permet d'imager les protéines avec une haute résolution en amplifiant la sensibilité de détection grâce au transfert de magnétisation. Ce projet vise à développer un outil de surveillance et de diagnostic de la maladie d’Alzheimer en imagerie non invasive, et à comprendre les mécanismes pathologiques des protéines mal repliées. Le laboratoire dispose d'une grande expertise en imagerie CEST, et des outils d'analyse avancés utilisant le machine learning seront développés pour quantifier les protéines mal repliées. Ce projet, mené au Centre de Recherche en Imagerie Moléculaire MIRCen-CEA à Fontenay-aux-Roses, offrira un nouvel outil non-invasif pour la recherche préclinique et améliorera la méthode CEST pour des applications cliniques. Le candidat devra maîtriser des outils de programmation et avoir des connaissances en RMN/IRM.

Proteinopathies, such as Alzheimer's, Huntington's, and Parkinson's diseases, are neurodegenerative disorders caused by misfolded proteins that aggregate in the brain, acquiring toxic functions and recruiting other soluble proteins. The aging population increases the importance of detecting and monitoring the aggregation of these proteins non-invasively for diagnostics and research. The MRI-CEST (Chemical Exchange Saturation Transfer) method allows imaging proteins with high resolution by enhancing detection sensitivity through magnetization transfer. This project aims to develop a non-invasive monitoring and diagnostic tool for Alzheimer's disease and to understand the pathological mechanisms of misfolded proteins. The laboratory has extensive expertise in CEST imaging, and advanced analysis tools using machine learning will be developed to quantify misfolded proteins. This project, conducted at the Molecular Imaging Research Center MIRCen-CEA in Fontenay-aux-Roses, will provide a new non-invasive tool for preclinical research and improve the CEST method for clinical applications. The candidate must be proficient in programming tools and have knowledge in NMR/MRI.
